蛋清源凝血酶抑制肽的纯化

Purification of anticoagulant peptides derived from egg white powder

摘要: 为了制备安全性高、副作用小的凝血酶抑制肽,将蛋清Alcalase酶解产物经凝胶柱色谱Sephadex G-50凝胶柱色谱和半制备高效液相色谱纯化,旋转蒸发、冷冻干燥后经酶标仪法检测其抗凝血活性,筛选出高抗凝血活性的组分。结果表明:经Sephadex G-50纯化后的第4个组分即洗脱时间为35~40 min的组分抗凝血活性最好,抑制率达到84.74%。将该组分通过半制备高效液相色谱纯化后得到12个峰,峰Ⅶ 即洗脱时间为17.275~19.542 min的组分活性最高,抑制率达到71.27%。

Abstract: The anticoagulant peptides derived from egg white powder were hydrolysised by Alcalase and then purified by SephadexG-50 and semi-preparative reversed phaseliquid chromatography (RPLC). High activity fraction was selected after rotary evaporation and freeze drying. The anticoagulant activity was determined by micro plate reader. The results showed that the No.4 fraction of eluted by SephadexG-50 at time 35~40 min had high anticoagulant activities which were 84.74%. Then, this fraction was purified by semi-preparative reversed phaseliquid chromatography in which it separated to 12 peaks. Peak Ⅶ was eluted at 17.275~19.542 min. The highest anticoagulant activities of fractions was peak Ⅶ which inhibition rate reached up to 71.27%.
