上皮-间质转化胶质瘤细胞的干细胞样特性及白藜芦醇对其干细胞样特性的抑制作用

Stem cell-like properties of glioma cells undergoing epithelial to mesenchymal transition and inhibitory effect of resveratrol on their stem cell-like properties

摘要: 目的:探讨发生上皮-间质转化(EMT)的胶质瘤细胞的干细胞样特性,阐明白藜芦醇(Res)对其干细胞样特性的抑制作用。方法:采用TGF-β1诱导胶质瘤U87细胞发生EMT,倒置显微镜观察细胞的形态表现,Western blotting法检测细胞间质标志物N-钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)和诱导EMT的转录因子(Snail、Slug、Zeb1和Twist1)表达水平。确定胶质瘤细胞发生EMT后,实验分为对照组(U87细胞不做处理)、EMT组(TGF-β1诱导胶质瘤细胞发生EMT)和Res处理组(Res处理发生EMT的U87细胞),检测各组胶质瘤U87细胞二代胶质球形成数和细胞中干细胞标志物(Bmi1和Sox2)表达水平。结果:经TGF-β1处理48 h的胶质瘤U87细胞表现出分散、拉长和类似成纤维细胞的外观。与对照组比较,不同浓度TGF-β1组胶质瘤细胞中N-cadherin、Vimentin、Snail、Slug、Zeb1和Twist1表达水平明显升高(P<0.05或P<0.01),表明胶质瘤U87细胞发生EMT改变。与对照组比较,EMT组胶质瘤U87细胞二代胶质球形成数明显增多(P<0.01),胶质瘤细胞中干细胞标志物Bmi1和Sox2表达水平升高(P<0.05或P<0.01)。与EMT组比较,Res处理组胶质瘤U87细胞二代胶质球形成数明显减少(P<0.01),胶质瘤细胞中干细胞标志物Bmi1和Sox2表达水平明显降低(P<0.01)。结论:发生EMT的胶质瘤U87细胞呈现干细胞样特性,Res能抑制其干细胞样特性。

Abstract: Objective: To investigate the stem cell-like properties of glioma U87 cells undergoing epithelial to mesenchymal transition (EMT), and to clarify the inhibitory effect of resveratrol (Res) on their stem cell-like properties.Methods: The glioma U87 cells were treated with transforming growth factor-β1(TGF-β1) to induce EMT, and the morphology of U87 cells was observed under inverted microscope. Western blotting method was used to detect the expression levels of mesenchymal markers (N-cadherin, Vimentin) and EMT-inducing transcription factors(Snail, Slug, Zeb1, Twist1). After the EMT of glioma cells was confirmed, the experiment was divided into control group (U87 cells didn't receive any treatment), EMT group (the glioma cells presented EMT induced by TGF-β1) and Res treatment group (the glioma cells with EMT were treated with Res). The number of second-generation gliospheres and the expression levels of stem cell markers (Bmi1 and Sox2) in glioma U87 cells in various groups were detected.Results: The glioma U87 cells treated with TGF-β1 for 48 h showed dispersed,elongated, and similar to the appearance of fibroblasts. The expression levels of N-cadherin, Vimentin, Snail, Slug, Zeb1 and Twist1 in glioma cells in different concentrations of TGF-β1 groups were significantly increased, suggesting the presence of EMT in glioma U87 cells. Compared with control group, the number of second-generation gliospheres in the U87 cells in EMT group was increased (P<0.01), and the expression levels of stem cell markers (Bmi1 and Sox2) in the glioma U87 cells in EMT group were markedly increased (P<0.05 or P<0.01). Compared with EMT group, the number of second-generation gliospheres in the glioma U87 cells in Res treatment group was significantly decreased(P<0.01), and the expression levels of Bmi1 and Sox2 in the glioma U87 cells in Res treatment group were decreased (P<0.01).Conclusion: The glioma U87 cells undergoing EMT exhibit the stem cell-like properties, and Res can inhibit their stem-like properties.
