三七总皂苷对再生障碍性贫血小鼠T淋巴细胞亚群的影响及其促造血作用

Influence of panax notoginseng saponins in T lymphocyte subsets in aplastic anemia mice and its hematopoietic effect

摘要: 目的:探讨三七总皂苷(PNS)对再生障碍性贫血模型小鼠T淋巴细胞亚群和造血功能的影响,阐明PNS对再生障碍性贫血小鼠的治疗机制。方法:50只小鼠随机分为对照组、模型组、低剂量PNS组、中剂量PNS组和高剂量PNS组,低、中和高剂量PNS组小鼠分别腹腔注射100、200和400 mg·kg-1PNS,对照组和模型组小鼠给予等体积生理盐水,连续给药14 d。测量小鼠体质量并计算脾脏及胸腺指数,采用流式细胞术检测小鼠外周血CD4+、CD8+T淋巴细胞百分率并计算CD4+/CD8+T淋巴细胞的比值,ELISA法测定小鼠血清γ干扰素(INF-γ)和肿瘤坏死因子α(TNF-α)水平,采用血细胞分析仪检测小鼠红细胞(RBC)、骨髓单个核细胞(BMCs)、白细胞(WBC)、血小板(Plt)数量及血红蛋白(Hb)水平。结果:与对照组比较,模型组小鼠体质量、脾脏指数和胸腺指数、外周血CD4+T淋巴细胞百分率及CD4+/CD8+T淋巴细胞比值明显降低(P<0.05),外周血RBC、BMCs、WBC、Plt数量和Hb水平明显降低(P<0.05),外周血CD8+T淋巴细胞百分率和血清中INF-γ、TNF-α水平明显升高(P<0.05)。与模型组比较,低、中和高剂量PNS组小鼠体质量、脾脏指数和胸腺指数、CD4+T淋巴细胞百分率和CD4+/CD8+T淋巴细胞比值明显升高(P<0.05),外周血RBC、BMCs、WBC、Plt数量和Hb水平明显升高(P<0.05),CD8+T淋巴细胞百分率和血清中INF-γ、TNF-α水平明显降低(P<0.05)。结论:PNS呈剂量依赖性调节T淋巴细胞亚群的平衡,抑制INF-γ和TNF-α释放,提高造血相关因子水平,从而改善再生障碍性贫血小鼠的造血功能。

Abstract: Objective:To investigate the influence of panax notoginseng saponins (PNS) in the T lymphocyte subsets and hematopoietic function in the aplastic anemia model mice, and to elucidate the therapeutic mechanism of PNS in the aplastic anemia mice. Methods:Fifty mice were randomly divided into control group, model group, low dose of PNS group, medium dose of PNS group, and high dose of PNS group.The mice in low,medium,and high doses of PNS groups were intraperitoneally injected with 100, 200, and 400 mg·kg-1PNS; the mice in control group and model group were intraperitoneally injected with normal saline of the same volume;lasted for 14 d. The body weights of the mice were measured and the spleen and thymus indexes were calculated after administration. Flow cytometry was used to detect the percentages of CD4+, CD8+T lymphocytes and the ratio of CD4+/CD8+T lymphocytes in the peripheral blood of the mice.The serum γ-interferon (INF-γ) and tumor necrosis factor-α(TNF-α) levels of the mice were determined by ELISA. The number of red blood cells (RBC), bone marrow mononuclear cells (BMCs), white blood cells (WBC), platelets (Plt) and the hemoglobin (Hb) level of the mice were measured by hematdogy analyzer. Results:Compared with control group, the body weight, the spleen index,the thymus index, the peripheral blood CD4+T lymphocyte percentage and the CD4+/CD8+T lymphocyte ratio of the mice in model group were significantly decreased (P<0.05); the number of RBC, BMCs, WBC, Pl tand the Hb level were significantly decreased(P<0.05); the peripheral blood CD8+T lymphocyte percentage and the serum INF-γ, TNF-α levels were significantly increased (P<0.05). Compared with model group, the body weights, the spleen indexes, the thymus indexes, the CD4+T lymphocyte percentages and the CD4+/CD8+T lymphocyte ratios of the mice in low, medium, and high doses of PNS groups were significantly increased (P<0.05); the number of BMCs, WBC, Plt and the level of Hb were significantly increased(P<0.05); the percentages of CD8+T lymphocytes and the levels of serum INF-γ and TNF-α of the mice were significantly decreased (P<0.05). Conclusion:PNS can improve the hematopoietic function of the aplastic anemia mice by regulating the balance of T lymphocyte subsets in a dose-dependent manner and inhibiting the release of INF-γ and TNF-α and increasing the levels of hematopoietic related factors.
