PCR Characters of a Recombined Tgo DNA Polymerase

摘要: 检测了重组Tgo酶的扩增性能、 耐热性以及长距离PCR能力, 结果表明, 该重组酶扩增性能与Taq酶相当; 其耐热性极强, 在 95 ℃ 40 min, 仍具有很强的聚合活性; 利用重组酶在常规PCR条件下, 成功扩增了5.3 kb的拟南芥Timeless基因片段.

Abstract: The amplification capability, the heat tolerance and the ability to long PCR of Tgo DNA polymerase were examined. The results reveal that Tgo DNA polym erase has the same amplification function asTaq enzyme but with a high fidelity. It also has a strong resistance to heat. Its amplification function would not be notably affected at 95 ℃ for 40 min. Via the recombinant enzyme, the 5.3 kb segment of Timeless gene was successfully amplified from Arabidopsis under the general conditions of PCR.
