摘要: 建立了一个快速、 准确、 灵敏的液相色谱串联质谱测定鼠血浆中莪术醇的分析方法. 血浆样品经乙腈沉淀蛋白后, 以甲醇-乙腈-10 mmol/L乙酸铵溶液(体积比: 42.5 ∶42.5 ∶15)为流动相, 采用反相色谱柱Nucleosil C18分离, 通过电喷雾离子化源的四极杆串联质谱仪, 以多反应监测方式检测. 线性范围为2.5~500.0 μg/L, 日间和日内精密度< 4.79%,平均提取回收率为100.7%.

Abstract: A rapid, simple and sensitive liquid chromatography/tandem mass spectro metry (LC/MS/MS) method was developed for the determination of curcumol in rat plasma. Samples of the plasma were deproteinated with acetonitrile, the analytes were then analyzed by reversedphase HPLC on a Hypersil ODS column with methanol-ace tonitrile-10 mmol/L ammonium acetate (volume ratio: 42.5 ∶42.5 ∶15) as mobile phase. Detection was carried out by multiple reaction monitoring on a API4000 LC/MS/MS system with an electrospray ionization (ESI) interface. The assay was linear in a range of 2.5~500.0 μg/L, intra and interday precisions over this range were < 4.79%, the average recovery was 100.7%.
