以线虫为模型考察中国芦荟提取物的降脂作用

Study on Effect of Aloe chinensis Baker Extract on Reducing Fat Accumulation by Using Model of Caenorhabditis elegans

摘要:

将鲜芦荟去皮研磨、 过滤, 先用磷酸盐缓冲液(PBS)进行提取、 稀释, 再配制含不同质量浓度D-葡萄糖的线虫生长培养基(NGM)喂食培养线虫. 以线虫寿命及运动能力等生理指标确定脂肪堆积模型的最佳糖诱导质量浓度; 通过油红O染色法及Image J软件分析评估芦荟提取物的降脂作用. 实验结果表明: 100 mL NGM中含    2 g D-葡萄糖为最佳糖诱导质量浓度, 可用于构建线虫脂肪堆积模型的脂肪评价; 芦荟提取物对线虫脂肪堆积模型有良好的降脂作用.

Abstract:

We peeled fresh Aloe chinensis Baker, extracted and diluted with PBS buffer. The nematodes were cultured on nematode growth medium containing with different mass concentrations of D-glucose, which was used to evaluate the optimal mass concentration by conventional physiological index such as lifespan, athletic ability of nematodes. And the effect of aloe extract on reducing fat accumulation was showed by oil red O staining and Image J software analysis. Experimental results show that the 2%(2 gD-glu/100 mL NGM) is the optimal induced mass concentration to establish the Caenorhabditis elegans fat accumulation model, and aloe extract has excellent effect on reducing fat accumulation model of C. elegans.
