HIV-GAG质粒DNA疫苗的纯化工艺

HIV-DNA Vaccine Research of Purification Technology

摘要:

采用中空纤维超滤浓缩、 离子交换介质和复合模式凝胶过滤介质纯化获得了HIV-GAG药用级质粒DNA疫苗, 并对菌体培养、 菌体裂解、 粗提分离和柱层析等环节进行研究. 实验结果表明, 产品基本符合药用级的质粒DNA要求.

Abstract:

In order to get HIVGAG pharmaceutical grade plasmid DNA vaccine, we studied  the procedure of the cultivation of bacteria, alkaline lysis, isolate rudely extraction and chromatography to get plasmid DNA. We used hollow fiber ultrafiltration concentration, ion exchange chromatography and composite model gel filtration medium in this study. The experimental results show that our final product meets the requirments of  pharmaceutical standardization. And
 this technology is suit for largescale production of DNA vaccines.
