摘要:
细胞分裂周期蛋白6(CDC6)是与多种细胞活动相关的三磷酸腺苷酶(ATPase)(AAA+ATPase)家族中的一员,具有ATPase活性,在真核生物中高度保守。CDC6是前复制复合体(pre-RC)的关键组成部分,参与启动DNA复制。同时还参与调控S/M期细胞周期检查点,调节细胞有丝分裂。CDC6在多种恶性肿瘤中异常表达,被视为是一种癌基因,其表达失调会导致DNA复制异常并发生DNA损伤,进而影响肿瘤的发生发展。多种药物和调节因子被证实可以抑制CDC6的表达,阻止肿瘤细胞的生长。因此,CDC6是一个潜在的治疗靶点和预后监测标志物。现基于国内外对CDC6的研究成果,对CDC6蛋白的结构、抑制剂及其在胶质瘤、前列腺癌(PCa)、乳腺癌和其他多种肿瘤中的发生机制进行综述,为恶性肿瘤的治疗提供参考。
