O-季铵化硫脲壳寡糖的制备及与鲱鱼精DNA的相互作

Preparation of OQuaternized Thiourea Chitooligosaccharidesand Interaction with Herring Sperm DNA

摘要: 以壳寡糖为原料, 制备O季铵化硫脲壳寡糖, 并用红外光谱、 紫外可见吸收光谱、 热重差热分析法对合成的化合物进行表征. 以鲱鱼精DNA为靶点, 用紫外可见吸收光谱、 循环伏安法和黏度法研究O-季铵化硫脲壳寡糖与鲱鱼精DNA的作用方式. 实验结果表明: O-季铵化硫脲壳寡糖在213.5,280.0 nm处出现吸收峰, 其热稳定性较差, 在180 ℃开始氧化分解; O-季铵化硫脲壳寡糖的紫外可见吸收峰在加入DNA后发生变化; O-季铵化硫脲壳寡糖使铁氰化钾DNA体系的氧化还原峰电流减小, 式量电位发生正移, 并使DNA的黏度增大, 即O季铵化硫脲壳寡糖与鲱鱼精DNA之间存在嵌插结合作用.

Abstract: O-quaternized thiourea oligosaccharides was prepared with chitosan oligosaccharides as raw materials, and its structure was characterized by infrared spectrum (IR), UVVis absorption spectra and thermogravimetric analysisdifferential thermal analysis (TG-DTA) methods. Using the herring sperm DNA as a target, the action mode of O-quaternized thiourea oligosaccharides and herring sperm DNA was studied by UV-Vis absorption spectra, cyclic voltammetry (CV) and viscosity method. The experimental results show that the absorption peak of Oquaternized thiourea oligosaccharides appears at 213.5 nm and 280.0 nm, its thermal stability is poor, and the oxygenolysis begins at 180 ℃. The UVVis absorption peaks of Oquaternized thiourea oligosaccharides change after adding DNA, O-quaternized thiourea oligosaccharides reduce the current of the redox peak of the potassium ferricyanideDNA system, potential shifts positively, and the viscosity of DNA increases. The action mode of O-quaternized thiourea oligosaccharides and DNA is intercalation.
