仿水溶剂促进酶促拆分环氧丙醇的研究

Enzymatic resolution of glycidol improved by water-mimicking solvent

摘要: 利用脂肪酶在非水介质中对外消旋环氧丙醇进行不对称酯合成反应, 重点研究了强极性有机溶剂作为仿水溶剂完全替代反应体系中的微量“必须”水对酶促拆分反应的影响. 实验结果表明, 乙腈作为仿水溶剂(最佳用量为0.4%)在适量无水硫酸钠(0.04 g左右)的配合下, 可以有效除去酯合成反应产生的水. 在最适反应条件下, 制备得到光学纯度约为90%的单一手性环氧丙醇酯.

Abstract: The asymmetric esterification of glycidol catalyzed by lipase was carried out in a nonaqueous medium. The effect of the replacement of water necessary for the funct ion of lipase with water-mimicking solvent on the resolution was investigated. It's found that the water produced from enzymatic esterification could be removed s uccessfully by addition of acetonitrile(0.4%, volume fraction) and 0.04 g anhydrous Na2SO4 into the reaction system. Under the optimal conditions, R- glycidyl butyrate with about ee 90% was prepared.
