四翅滨藜LRR类受体蛋白激酶基因在酵母中的表达

摘要:

从四翅滨藜(Atriplex canescens)cDNA文库中得到一个亮氨酸富集重复序列类受体蛋白激酶(Leucinerich repeat receptorlike protein
 kinases, LRRRLKs)的全长cDNA序列, 命名为AcLRR(登录号: JN974247). 为研究AcLRR的抗逆功能, 将其构建到酵母表达载体pYESDEST52, 并转化到酿酒酵母INVSc1菌株中, 获得重组酵母INVSc1(pYESAcLRR), 对其进行盐、 碱、 干旱、 高温、 冷冻和活性氧等胁迫, 分析在不同逆境胁迫下的表型特征. 实验结果表明, INVSc1(pYESAcLRR)在各种胁迫下的存活率明显高于对照, 表明AcLRR基因具有抗NaCl,KCl,NaHCO3,Na2CO3、 干旱、 高温、 冷冻和活性氧等胁迫的能力, 特别对冷冻和活性氧表现出明显抗性. 由此可推测该基因参与了四翅滨藜的抗逆调控过程.

Abstract:

A full length leucinerich repeat receptorlike protein kinase (LRR-RLK) was obtained from a fulllength Atriplex canescens cDNA library, named as
AcLRR (GenBank accession number: JN974247). To investigate the stress tolerance of AcLRR, the recombinant plasmid pYES-AcLRR was constructed by inserting the AcLRR gene into the yeast expression vector pYES\|DEST52 via LR recombination. The recombinant plasmid pYES-AcLRR that was transformed into yeast Saccharomyces cerevisiae INVSc1, and the INVSc1 strain transformed with empty pYESDEST52 was used as the control. The recombinant yeast INVSc1 harboring pYES-AcLRR and the control INVSc1 harboring empty pYESDEST52 were treated with salinealkali, drought, high temperature, frezzing and paraquat, respectively, and their survival rates were compared under these stress conditions. The results show that the survival rates of the recombinant yeast INVScl (pYES-AcLRR) were higher than those of the control strain under these stress conditions, indicating that the AcLRR was tolerant to NaCl,KCl,NaHCO3,Na2CO3, drought, high temperature, frezzing stresses, which exhibits that AcLRR may play an important role in stress tolerance of A.canescens.
