Determination of Curcumol Concentration in Rat Plasma by Liquid Chromatographytandem Mass Spectrometry

Abstract: A rapid, simple and sensitive liquid chromatography/tandem mass spectro metry (LC/MS/MS) method was developed for the determination of curcumol in rat plasma. Samples of the plasma were deproteinated with acetonitrile, the analytes were then analyzed by reversedphase HPLC on a Hypersil ODS column with methanol-ace tonitrile-10 mmol/L ammonium acetate (volume ratio: 42.5 ∶42.5 ∶15) as mobile phase. Detection was carried out by multiple reaction monitoring on a API4000 LC/MS/MS system with an electrospray ionization (ESI) interface. The assay was linear in a range of 2.5~500.0 μg/L, intra and interday precisions over this range were < 4.79%, the average recovery was 100.7%.
