Determination of Metabolites of Anthraquinone in Rat Plasma and Urine by LC-MS

Abstract:

To establish a method to identify the metabolites of anthraquinones in the plasma and urine of rats  administrated Polygonum cuspidatum extract
 by liquid chromatographymass spectrometry (LC-MS). The assay was conducted on an Agilent XDBC18 (4.6 mm×250 mm, 5 μm), the mobile phase was watermethanol gradient elution, the column temperature was 25 ℃, the ion source was ESI in negative mode and capillary voltage was 3.5 kV. Three metabolites, emodin, chrysophanol and physcion were determined in rat plasma. Four metabolites emodin, emodin glucuronic acid ester, Aloeemodin sulfate and aloeemodin glucuronic acid ester were determined in rat urine.
