聚乳酸/羟基乙酸胆总管支架在体外胆汁中的降解

Degradation of polylactic acid/glycolic acidstents in bile in vitro

摘要: 目的:观察不同质量比聚乳酸/羟基乙酸(PLGA)胆总管支架在体外胆汁中质量、相对分子质量及外形变化的情况,找出在胆总管探查术中可替代T管作用的理想质量比的支架。方法:选用不同质量比(50/50、60/40、70/30和80/20)的PLGA可降解支架,每种比例各45只,放入装有新鲜人体胆汁的安瓶中,置于37℃恒温培养箱,每日更换胆汁,隔日观测 支架外形、质量和相对分子质量的变化。结果:50/50和60/40配比的支架降解高峰在9~10 d;70/30配比的支架降解高峰在13~14 d;80/20配比的支架降解高峰在17~18 d,符合临床胆总管探查术后T管拔除的时间(一般为14 d)。结论:80/20配比的PLGA可降解支架的降解高峰在临床T管拔除时间之后,可替代T管的作用。

Abstract: Objective To study the changes of different mass-ratio of polylactic acid/glycolicacid (PLGA) common bile duct stent in bile in vitro, including mass, relative molecular mass as well as physical appearance in order to find an ideal substitute for T tube in common bile duct exploration.Methods 45 stents of four different ratios (50/50, 60/40, 70/30 and 80/20) of PLGA were used respectively. All the stents were submerged into the vials with fresh human bile and incubated at 37℃. The bile was changed every day and the changes of mass, relative molecular mass and physical appearance were observed every other day. Results The degradation peak times of 50/50 and 60/40 ratio was 9-10 d, that of 70/30 stents was 13-14 d, and 80/20 ratio was 17-18 d. The degradation peak time of 80/20 ratio was consistent with the termination time(14 d ) of T tube.Conclusion The degradation time of 80/20 ratio stents is consistent with the termination time of T tube, T tube can be replaced by this ratio stent.
