抗诱变中草药的正交实验

摘要: 目的:通过正交实验确定抗诱变中草药人参、五加皮、猪苓和甘草保护染色体稳定性的最佳配伍方案。方法:采用徐道觉(Hsu TC)建立的以博来霉素(BLM)为诱变剂的诱变剂敏感性实验方法,将上述4味中草药药量取3个水平:大剂量(常用量2倍)、常用剂量及零水平;把此4味中药随机分组,人参: C、五加皮:D、猪苓:A、甘草:B;分别在培养的人外周血淋巴细胞中加入不同剂量配比的人参、五加皮、猪苓和甘草,比较各剂量配比中草药干预后的染色体断裂率(b/c:每分裂相染色体的断裂数),观察它们的抗诱变效果。 结果:人参、五加皮、甘草和猪苓的极差分别为16、14、 9和6,配伍中4味中草药主次顺序依次为人参、五加皮、猪苓、甘草;最佳配比A1B2C1D2。 结论:采用最佳染色体稳定剂配比方案(A1B2C1D2)进行药物配伍可以最大限度地抵抗诱变剂BLM的作用,增强染色体的稳定性。

Abstract: Objective To obtain the best compatibility prescription to protect the stability of chromosome in which the Chinese herbal medicines had the role of anti-mutagesis including ginseng,slenderstyle acanthopanax root-bark, agaric, and Glycyrrhiza uralensis with orthogonal experiment. Methods The mutagen-sensibity experiment created by Hsu TC in which Bleomycin (BLM) was used as mutagen was adopted. The dosages of the above-mentioned Chinese herbal medicines in the experiment were applied in 3 levels: large dosage (double commonly used dosage), commonly used dosage and zero. The traditional Chinese medicines were randomly divided into four groups:A,B,C, and D. Ginseng was represented by C; slenderstyle acanthopanax root-bark was represented by D; agaric by A and Glycyrrhiza uralensis by B. The Chinese herbal medicines composed of different ratio with ginseng,slenderstyle acanthopanax root-bark, agaric, and Glycyrrhiza uralensis,respectively, were added into cultured lymphocytes of human peripheral blood.The chromosome breakage rates (b/c: chromosome breakage number per cell) intervened by the Chinese herbal medicines composed of different dosage ratio were compared and the anti-mutagesis effects were observed. Results The ranges of ginseng,slenderstyle acanthopanax root-bark ,agaric, and Glycyrrhiza uralensis was 16,14,9, and 6,respectively;the importance of the 4 kinds of Chinese herbal medicines in the compatibility prescription in order was ginseng,slenderstyle acanthopanax root-bark, agaric, and Glycyrrhizia uralensis; the best compatibility prescription was A1B2C1D2. Conclusion The best compatibility prescription to protect the stability of chromosome is A1B2C1D2.It is the greatest resistance to anti-mutagenic agents and reinforcement of chromosome stability to adopt above-mentioned dosage ratio.
