开博通对局灶性脑缺血大鼠MMP-2及TIMP-2的影响

Effects of Captopril on MMP-2 and TIMP-2 after focal cerebral ischemia in rats

摘要: 目的:观察大鼠大脑中动脉(MCA)缺血后MMP-2及TIMP-2的动态变化及开博通对局灶性脑缺血大鼠的脑保护作用。 方法:采用线结扎颈总动脉加缺氧诱导的方法制作大鼠MCA局灶性脑缺血模型,免疫组织化学法测定各组大鼠脑组织中MMP-2及TIMP-2的表达。 结果:缺血5 d,海马、基底及皮质MMP-2表达明显增加(P<0.05);开博通可抑制MMP-2的表达(P<0.05),具有脑保护作用。开博通对TIMP-2作用不明显。 结论:MMP-2加重缺血性脑损伤,基质金属蛋白酶抑制剂开博通具有重要的脑保护作用。

Abstract: Objective To observe the dynamic changes of MMP-2 and TIMP-2 after middle cerebral artery (MCA) ischemia in rats and evaluate the neuroprotective effects of Captopril on focal cerebral ischemia. Methods The rat focal cerebral ischemia models were made by using ligating the middle cerebral artery. The changes of MMP-2 and TIMP-2 in rat brain tissues in different groups were measured by immunohistochemical method. Results The MMP-2 expression was significantly incresed in the ischemia regions 5 d after inschemia (P<0.05), and Captopril could decrease the expression of MMP-2 (P<0.05); there were no differences of the effects of Captopril on TIMP-2 between various groups. Conclusion MMP-2 can aggravate cerebral damage after ischemia,and Captopril, an artificial inhibitor of metalloproteinase, plays an important protective role in brain.
