MMP-9及其mRNA在胃癌间质血管壁的表达

摘要: 目的:观察基质金属蛋白酶9(MMP-9)及其mRNA在胃癌间质中的表达情况及其与肿瘤浸润、转移及间质血管新生的关系。 方法:利用74例胃癌标本,经连续切片免疫组织化学染色、原位杂交及形态定量分析法,进行MMP-9、MMP-9 mRNA、CD34及α-SMA的检测,并结合临床病理参数进行综合分析。 结果:MMP-9及其mRNA的表达均与癌组织的浸润深度、分化程度、淋巴结转移有关;间质微血管密度 (MVD)与癌组织的浸润深度、淋巴结转移有关;24例MMP-9表达于血管内皮细胞和周细胞:4例间质毛细血管内皮细胞MMP-9 mRNA阳性,其中3例血管壁周细胞MMP-9 mRNA阳性。 结论:胃癌细胞自身具有产生与分泌MMP-9的能力,MMP-9对肿瘤的侵润、转移及间质血管新生具有促进作用。

Abstract: Objective To investigate the expressions of MMP-9 and MMP mRNA in gastric carcinoma interstitium and the relationships between the expressions and the invasion,metastasis and angiogenesis of the neoplasm. Methods MMP-9,MMP-9 mRNA, CD34, α-SMA in 74 cases of gastric carcinoma were examined by immunohistochemistry, in situ hybridizati on and morphometric analysis of secquential slides. The relationships between the expressions of MMP, MMP mRNA and the clinical parameters were analysed. Results The expressions of MMP-9 and MMP mRNA were closely related to the invasion, differentiation and lymph node status of the gastric carcinoma. The microvessel density (MVD) had great correlation with the invasion and lymph node status of the carcinoma. Endothelia and pericytes were MMP-9 positive in 24 cases.Endothelia were MMP-9 mRNA positive in 4 cases among which pericytes of 3 cases were MMP-9 mRNA positive. Conclusion Gastric carcinoma cells have the ability to produce and secret MMP-9 which can promote the invasion,metastasis and angiogenesis of the neoplasm.
