抑菌中药银花泌炎灵片的体外和体内抗细菌内毒素作用

Anti-lipopolysaccharide effect of antibacterial traditional Chinese medicine Yinhuamiyanling Tablet in vivo and in vitro

摘要:

目的:探讨传统抑菌中药银花泌炎灵片的抗细菌内毒素(LPS)作用,为其应用提供理论依据。方法:以银花泌炎灵片为研究对象,体外采用鲎实验法检测其抗LPS活性。体内实验选取90只ICR小鼠,随机分为空白对照组,药物对照组,LPS对照组,LPS低、中、高剂量预防组和治疗组,每组10只。低、中、高剂量预防组和治疗组小鼠分别灌服低剂量(0.4 g/mL)、中剂量(0.5 g/mL)和高剂量(0.6 g/mL)银花泌炎灵片溶液,药物对照组小鼠灌服0.6 g/mL银花泌炎灵片溶液,LPS对照组和空白对照组小鼠分别灌服等体积生理盐水。除空白对照组和药物对照组外,各预防组(于末次给药后3h)、治疗组(于第1次给药前3 h)和LPS对照组均按18 mg/kg腹腔注射LPS溶液,记录给予LPS溶液后48 h内各组小鼠的体质量、血液指标和脏器指数。结果:体外鲎实验法,将100 g/L银花泌炎灵溶液稀释至6.25 g/L后仍然具有抗LPS活性。体内实验,与空白对照组比较,LPS对照组小鼠体质量、白细胞(WBC)数、红细胞(RBC)数、血红蛋白浓度(Hb)、红细胞比容(Hct)和血小板(PLT)数均降低(P<0.05或P<0.01),脾脏和肺脏指数升高(P<0.01)。与LPS对照组比较,低剂量治疗组和中剂量预防组小鼠体质量在注射LPS后24 h升高(P<0.05),高剂量和中剂量预防组小鼠体质量在注射LPS后36 h升高(P<0.05),中剂量预防组小鼠体质量在注射LPS后48 h升高(P<0.05);高剂量治疗组小鼠WBC数升高(P<0.05);低剂量治疗组和中剂量预防组小鼠RBC数升高(P<0.01),低剂量治疗组和低、中、高剂量预防组小鼠Hb数升高(P<0.05或P<0.01),低、高剂量预防组小鼠Hct数升高(P<0.05或P<0.01),低、高剂量预防组小鼠PLT数升高(P<0.05);中剂量治疗组小鼠肝脏指数降低(P<0.01);高剂量治疗组小鼠肺脏指数降低(P<0.05);低、高剂量治疗组和低、中、高剂量预防组小鼠脾脏指数均显著降低(P<0.05或P<0.01)。结论:银花泌炎灵片在体外和体内均具有抗细菌LPS的活性。

Abstract:

Abstract:Objective
To discuss the anti-lipopolysaccharide(LPS) effect of the traditional antibacterial Chinese medicine Yinhuamiyanling Tablet,and to provide theoretical basis for its application. Methods Yinhuamiyanling Tablet were regarded as the study object,and the limulus test method was used to detect the anti-lipopolysaccharide activity.In vivo tests,90 ICR mice were divided into blank control group,drug control group,LPS control group,and low dose,middle dose,high dose prevention and treatment groups,and there were 10 mice in each group.The mice in low,middle,and high dose prevention and treatment groups were respectively fed with low dose (0.4 g/mL),middle dose (0.5 g/mL),high dose (0.6 g/mL) of Yinhuamiyanling Tablet solution.The mice in drug control group were fed with 0.6 g/mL  Yinhuamiyanling Tablet solution,and the mice in LPS control group and blank control group were fed with an equal volume of saline.In addition to  blank control group and drug control group,the mice in different prevention groups (3 h after the last administration) and different treatmentgroups(3 h before the first  administration) and LPS control group were intraperitoneally injected with 18 mg/kg LPS solution.48 h after LPS injection,the changes of body weight,blood parameters and organ indexes of the  mice in each group were observed.
Results The limulus test in vitro results showed that Yinhuamiyanling Tablet solution had anti-bacterial LPS activity even though its concentration was diluted to 6.25 g/L from 100 g/L.In vivo, compared with blank control group,the body weight,the number of white blood cells (WBC),red blood cells (RBC),concentration of haemoglobin(Hb),hematocrit(Hct),platelet(PLT) of the mice in LPS control group were significantly decreased (P<0.05 or P<0.01);the spleen indexes and lung indexes were significant increased(P<0.01).Compared with LPS control group,the body weight of the mice in  low dose treatment group and middle dose prevention group were significantly increased 24 h after LPS injection(P<0.05);the body weight of the mice in  middle dose and high dose prevention groups were significantly increased 36 h after LPS injection(P<0.05);the body weight of the mice in middle dose prevention groups was significantly increased 72 h after LPS injection(P<0.05);the number of WBC of the mice in high dose treatment group was significantly increased (P<0.05),the number of RBC of the  mice  in low dose treatment group and middle dose prevention group was significantly increased  (P<0.01);the number of  Hb of the mice in low dose treatment group and different prevention groups was significantly increased (P<0.05 or P<0.01);the number of Hct of the  mice in low dose and high dose prevention groups was significantly increased (P<0.05 or P<0.01);the number of PLT of the mice in low and  high dose prevention groups was significantly increased  (P<0.05);the liver index of the  mice in  middle dose treatment group was significantly decreased (P<0.01);the lung index of the mice in high dose treatment group was significantly decreased (P<0.05);the spleen indexes of the mice in low dose,high dose treatment groups and different  prevention groups was significantly deceased (P<0.05 or P<0.01).Conclusion Yinhuamiyanling Tablet has anti-bacterial LPS activity in vitro and in vivo.
