rhKD/APP对急性肝损伤小鼠的保护作用

Hepatoprotective effects of rhKD/APP on acute hepatic injury 

摘要: 目的:观察rhKD/APP对四氯化碳(CCl4)所致急性肝损伤的保护作用,探讨其可能的作用机制。方法:采用CCl4建立小鼠急性肝损伤模型,取小鼠60只随机分为6组(每组10只),分别设为空白对照组、模型组、阳性药对照组及rhKD/APP小中大剂量(4.5、9.0、18.0 mg•kg-1)组。测定实验小鼠血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)及肝组织丙二醛(MDA)、肝重系数并观察肝脏组织病理学变化。结果: rhKD/APP小、中、大剂量组血清ALT、AST活性和肝组织中MDA低于模型组(P<0.05或P<0.01);肝脏组织病理切片显示,模型组有炎细胞浸润,肝细胞坏死,与模型组相比,rhKD/APP小、中、大剂量组肝组织损伤程度较轻,且随着rhKD/APP剂量增加肝组织损伤程度逐渐减轻。结论:rhKD/APP对CCl4所致急性肝损伤小鼠具有明显的保护作用。

Abstract: To observe the protective effects of rhKD/APP on carbon tetrachloride-induced acute hepatic injury and find possible mechanisms of hepatic injury. Methods Sixty mice were randomly divided into 6 groups(n=10):control,model,pos itive control and rhKD/APP groups with different doses (4.5,9.0,18.0 mg•kg -1). Pathologic models of acute hepatic injury in mice caused by carbon tetrachloride were set up and rhKD/APP with different dosage were administered (ip) to mice.Effects of rhKD/APP on hepatic injury were observed by measuring enzyme level (AST and ALT)in serum,hepatic malondialdehyde and morphology of hepatic tissue section . Results Compared with model group, serum ALT and AST concentrations in rhKD/APP groups with different doses were decreased significantly (P<0.01, P<0.05), MAD in hepatic tissue in rhKD/APP groups with different doses were decreased (P<0.01, P<0.05). The pathological results showed that there were inflammatory cells and hepatocellular necrosis in model group, the protective effects of rhKD/APP on hepatic injury showed linear relation to the drug dose. Conclusion rhKD/APP has markedly protective effects on acute hepatic injury induced by carbon tetrachloride.
