N-甲基-D-天门冬氨酸诱导Wistar乳鼠癫痫发作与West综合征动物模型的比较

Comparison between seizures induced by N-methyl-D-Aspartatein Wistar rats and animal model of West syndrome

摘要: 目的:探讨建立West综合征动物模型的可能性。方法:选取12日龄Wistar乳鼠(n=64),其中32只随机分成N-甲基-D-天门冬氨酸(NMDA)处理组(n=16)和对照组(n=16),连续14 d腹腔注射NMDA或生理盐水,观察有无癫痫发作并对其痫性发作的类型、潜伏期、发病率及行为改变均做以相应记录;其余12日龄Wistar乳鼠分NMDA处理组(n=16)和对照组(n=16),分别进行皮质脑电图变化记录。结果:对照组无癫痫发作,12~13日龄Wistar乳鼠引出甩尾等自动症及前弓反张发作,14~25日龄Wistar乳鼠只有自动症而没有前弓反张发作。对照组皮质脑电图无明显癫痫性波形发放;NMDA处理组则在自动症及癫痫发作期间出现尖波、棘波等特征性改变。结论:NMDA诱导Wistar乳鼠产生一种独特的、年龄依赖性癫痫发作,虽未满足West综合征动物模型的全部标准,但发作表现和脑电图改变与儿童时期West综合征有极为相似之处。

Abstract: Objective To explore the possibility of establishment of the animal model of West syndrome. Methods Experiment was performed on 12-day-old Wistar strains (n=64). 32 among them were randomly divided into N-methyl-D- Aspartate (NMDA) treated group(n=16)and control group(n=16), and had an intraperitoneal injection of NMDA or NS respectively for 14 d in succession. The behavioral manifestations and/or the seizures induced by NMDA in each group were observed and the incidence, latencies, pattern of motor seizures were noted. The changes of ECoG were observed in the rest of Wistar strains (n=32) which were divided into NMDA treated group (n=16) and control group(n=16). Results The control group had no seizures. NMDA-induced automatisms and emprosthotonus in (12-13)-day-old rats were elicited while automatisms were only elicited by NMDA from 14-day-old rats to 25-day-old rats without emprosthotonus. There were no obvious epileptiform discharge in control rats compared with the NMDA-treated rats which had the characteristic changes of shape waves and spike waves concurrently during automatisms and emprosthonotonic seizures. Conclusion Although the unique seizures with age-specificity induced by NMDA in developing rats can not fulfills all criteria of the West syndrome model, it partially resembles West syndrome in childhood on the aspects of behavioral manifestations and ECoG changes.
