摘要: 目的:探讨天然产物虎眼万年青多糖(S3)对T细胞及其细胞因子IL-2 mRNA表达的影响,进一步从细胞和分子水平探讨其免疫调节作用机制,为开发这一天然产物提供理论依据。 方法:利用流式细胞术,检测S3对T淋巴细胞亚群CD3、CD4、CD8、CD4/CD8阳性细胞百分率的影响,并采用RT-PCR方法检测IL-2 mRNA的表达水平。 结果:各剂量组CD3、CD4阳性细胞百分率均有上升趋势,极高剂量组CD3、CD4与对照组比较差异有显著性(P<0.001),CD8有下降趋势,各剂量组均能显著提高CD4/CD8阳性细胞百分率(P<0.001);极高剂量组和高剂量组可明显促进脾细胞中细胞因子IL-2的mRNA表达,使表达量增加(P<0.01,P<0.05)。 结论:S3具有较强的增强机体多种免疫功能的作用。

Abstract: Objective To study the effects of the polysaccharide (S3) separated out in 60% alcohol aqueous from Ornithogalum caudatum ait (OCA) on the expressions of T cells and IL-2 mRNA. Methods After separation of the lymphocytes from lymphoid nodes of BALB/c mouse, the effects of S3 on the expression rates of CD3, CD4, CD8, and CD4/CD8 on T cells were determined by flow cytometry following two-colour inmmunofluorescent staining. The expressions of IL-2 mRNA were also detected by RT-PCR. Results S3 promoted the positive proliferation of CD3, CD4, CD4/CD8 and made CD8 declined among the T cell lines. RT-PCR analyses showed that mRNA expressions of IL-2 in splenocytes could be promoted in both the extremely high dose and the high dose groups (P<0.05, P<0.01, respectively). Conclusion S3 possesses the effects to promote immune functions in mice.
