地塞米松对内毒素休克大鼠脑皮质TLR4、IκBα和IL-18 mRNA表达的影响

Effects of Dex on TLR4,IκBα and IL-18 mRNA expressions of cortex in endotoxic shock rats

摘要: 目的:通过观察内毒素休克大鼠脑皮质中TLR4、IκBα和IL-18 mRNA的表达及地塞米松(Dex)对其的影响,探讨内毒素引起脑组织损伤的分子机制。方法:18只Wistar大鼠随机分为实验对照组(Control)、内毒素休克组(LPS)和地塞米松组(LPS+Dex),每组6只。除对照组外,大鼠静脉注射内毒素(4 mg•kg-1),对照组大鼠静脉注射等量葡萄糖溶液;4 h后检测脑皮质中TLR4、IκBα和IL-18 mRNA的表达。结果:LPS+Dex组平均动脉压明显高于LPS组(P<0.01),大鼠存活率亦明显高于LPS组(P<0.01)。LPS+Dex组TLR4、IL-18mRNA表达均明显低于LPS组(P<0.05),而IκBα mRNA表达明显高于LPS组(P<0.05)。 结论:Dex能下调脑组织中TLR4 mRNA表达,而上调IκBα mRNA表达,对中枢神经系统具有一定的保护作用。

Abstract: Objective To explore the molecular mechanism of brain tissue injury induced by endotoxin and observe the effects of Dex on TLR4,IκBα and IL-18 mRNA expressions of cortex in the endotoxic shock rats.Methods Eighteen Wistar rats were randomly divided into LPS,LPS+DEX and control group(n=6).Except control group,all the rats were administered with LPS(4 mg•kg-1) by intravenous injection.The rats in control group were given glucose solution at same volume.The TLR4,IκBα and IL-18 mRNA expressions were assayed 4 h after intravenous injection.Results The mean arterial pressure (MAP) and the survival rate in LPS+DEX group were significantly higher than those in the LPS group(P<0.01).TLR4 and IL-18 mRNA expressions in LPS+DEX groups were obviously lower than those in LPS group (P<0.05).The IκBα mRNA expression in LPS+DEX group was significantly higher than that in LPS group (P<0.05).Conclusion Dex may down-regulate TLR4 expression and up-regulate IκBα expression in the brain tissue,and has protective effect on central nervous system.
