摘要: 目的:观察基质金属蛋白酶9(MMP-9)蛋白及其mRNA在胃癌组织与胃癌细胞BCG823中的表达特点,并探讨其对胃癌发展的作用。 方法:采用免疫组织化学染色、原位杂交、侵袭实验及形态定量分析方法,对74例胃癌组织标本及体外培养的胃癌细胞BCG823,进行MMP-9、MMP-9 mRNA的检测,并结合临床病理参数进行综合分析。 结果:MMP-9蛋白及其mRNA的表达均与癌组织的浸润深度、分化程度、淋巴结转移密切相关;BCG823细胞 MMP-9蛋白及mRNA染色均为阳性;MMP-9抗体可使肿瘤细胞的体外侵袭、移动能力下降。 结论:胃癌细胞自身具有产生与分泌MMP-9的能力,后者对肿瘤的浸润、转移具有促进作用。

Abstract: Objective To investigate the expressions of MMP-9 protein and MMP-9 mRNA in the tissues and cells of the gastric carcinoma and the effects of MMP-9 on the cancer progress. Methods The expressions of MMP-9 and MMP-9 mRNA in 74 cases and the BCG823 cells of gastric carcinoma cultured in vitro were examined by immunohistochemistry, in situ hybridzation, invasion test and morphometric analysis. The relationships between the expressions and the clinical parameters of gastric carcinoma were analysed. Results The expressions of MMP-9 and MMP-9 mRNA were closely related to the invasion, differentiation, and lymph nodes status of the gastric carcinoma. The MMP-9 and MMP-9 mRNA were positive in BCG823 cells. The antibody to MMP-9 could decrease the invasion and metastasis ability of tumor cells. Conclusion Gastric carcinoma cells have the ability to produce and secrete MMP-9 which can promote the invasion and metastasis of the neoplasm.
