rhKD/APP对大鼠急性胰腺炎血清炎性因子和胰腺组织超微结构的影响

Effects of rhKD/APP on inflammatory cytokines in serum and ultrastructrue of tissue in acute pancreatitis rats

摘要: 目的:观察rhKD/APP对实验性大鼠急性坏死性胰腺炎(ANP)血清炎性因子和胰腺组织超微结构的影响。方法: 采用3.5 %的牛磺胆酸钠0.1 mL•100g-1 逆行注入大鼠胰胆管内诱导ANP模型。60只大鼠随机分为假手术组(Sham组),模型组(Model组),rhKD/APP低、中、高剂量组(4×104 kU•kg-1、8×104 kU•kg-1、16×104 kU•kg-1组),阳性药抑肽酶组(Aproitin组),观察ANP大鼠血清肿瘤坏死因子-α (TNF-α) 、一氧化氮 (NO)变化和胰腺组织超微结构变化。 结果:与假手术组比较, 模型组大鼠血清TNF-α和NO水明显升高(P< 0.001); 与模型组比较,rhKD/APP中、高剂量组大鼠血清TNF-α和NO均降低(P< 0.05或P< 0.01),与阳性药抑肽酶组比较差异无显著性。胰腺组织超微结构显示细胞表面可见较多微绒毛,核仁明显,分泌颗粒增多。结论:rhKD/APP早期给药可显著减轻胰腺炎症反应,对大鼠急性胰腺炎具有治疗作用。

Abstract: To observe the effect of rhKD/APP on inflammatory cytokines and ultrastructrue in experimental acute necrosis pancreatitis(ANP) in rats.Methods ANP model was induced by injections of 3.5 % sodium taurocholate into the main pancreaticduct of rats.Sixty rats were randomly divided into 6 groups(n=10):sham group;model group;low,mediate,high dose rhKD/APP groups(4×104 ,8×104 and 16×104 kU•kg-1);aproitin group.TNF-α and N activity wereassayed and ultrastructrue changes were observed after treatment with rhKD/APP.Results Compared with sham group,the levels of TNF-α and NO in serum in model group rapdily raised(P<0.01).Compared with model group,the levels of serum TNF-α and NO in mediate and high dose rhKD/APP groups were decreased(P<0.05 or P<0.01);but there was no significant difference compared with aproitin group.The ultrastructure results of pancreas tissue showed that there were a lot of microvilli on cell surface,the nucleoli were significant,and secretory granules inreased. Conclusion rhKD/APP can reduce the inflammatory response of pancreas,and has the rapeutic effect on rats with ANP.
