Neuroprotective effects of soy isoflavones on PC12 cells based on antioxidation

Abstract: Experiment results show that when PC12 cells were exposed to 20.00 μg/mL genistein (GEN), daidzein (DAI) or their combination (GEN-DAI), the cells' viability rates, mophologies, mitochondrial membrane potential, superoxide dismutase activities and inhibition rates were not changed. However, different changes of PC12 cells were observed under different treatments. The reactive oxygen species level, the relative fluorescence intensity and malondialdehyde of PC12 cells were increased by GEN. The leakage rate of lactic dehydrogenase was decreased in DAI and in GEN-DAI combination treatments, the glutathione contents were increased to (297.27±9.47) μmol/(g·prot) and (269.90±00) μmol/(g·prot) respectively. The acetylcholine content in culture medium and metabolism of cholinergic system was increased in GEN-DAI treatment. GEN-DAI combination treatment can more significantly increase the activity of the antioxidant system of PC12 cells than GEN and DAI treatments.
