平板扩散法测定黄嘌呤氧化酶的活性

摘要:

通过对Arthrobacter- sp.X11菌株所产黄嘌呤氧化酶(XOD)活性测定方法的研究和比较, 建立一种简便可行的黄嘌呤氧化酶活性快速检测方法, 即平板扩散法. 实验结果表明, 该方法测得的显色圈直径与辣根过氧化物酶分光光度法测得的酶活力之间有良好的线性关系, 通过测量显色圈的直径即可估算出样品的酶活力.

Abstract:

Compared with spectrophotometry for the determination of xanthine oxidase activity by horseradish peroxidase, a quick and simple solid agar plate method of assaying the activity of xanthine oxidase(XOD) produced by Arthrobacter sp.X11 was developed with xanthine as a substrate. The activity of xanthine oxidase was analyzed quickly and roughly according to the diameters of color haloes. This method was used to optimize nitrogen source in a fermentation medium for enzyme production.
