薄荷提取物对提高秀丽线虫应激抵抗能力的影响

Effect of Mint Extracts on  Enhancing Stress Resistance of Caenorhabditis elegans

摘要: 以秀丽线虫为模型, 研究药食同源植物薄荷提取物对其应激抵抗能力的影响, 并以紫外辐射为应激条件, 进一步研究薄荷提取物抵抗紫外辐射的作用机制.  结果表明: 薄荷提取物可明显提高线虫抵抗紫外辐射、 绿脓杆菌感染、 35 ℃热应激和胡桃醌氧化的应激能力; 薄荷提取物可明显降低紫外辐射后线虫体内活性氧(ROS)水平、提高超氧化物歧化酶(SOD)活性,并可通过胰岛素/IGF-1样信号通路发挥作用,  提高通路下游转录因子sod-3,hsp-12.6,hsp-16.1和hsp-16.49的mRNA水平表达量以及转录因子daf-16的细胞核定位数量;  薄荷提取物可提高线虫的运动能力和吞咽能力, 但不影响其寿命和生育能力.

Abstract: Taking Caenorhabditis elegans (C.elegans)  as a model, we studied the effect of mint extracts on the stress resistance, and further studied the mechanism of mint extract against ultraviolet (UV) irradiation. The results show that  mint extracts can significantly increase the resistance of C.elegans to  UV-irradiation,P.Aeruginosa infection, 35 ℃ heat shock,  and juglone oxidation. The mint extracts can significantly reduce the level of reactive oxygen species (ROS)  and increase activity of superoxide dismutase (SOD) in C.elegans   after UV-irradiation. It can also play a role  through insulin/IGF-1-like signaling pathway, increase the mRNA expression  levels of the sod-3,hsp-12.6,hsp-16.1, and hsp-16.49  of the downstream transcription factor of the pathway, and increase the number of nuclear localization of transcription factor daf-16. The mint extracts can increase the motility and swallowing ability of C.elegans, but do not affect their lifespan and fertility.
