Statistical Optimization of FermentationMedium for Nisin Production

Abstract:

The PlackettBurman design was used to evaluate the effects of eight variables which could affect the nisin production. On the basis of analyzing the statistical regression, yeast extract, glucose and K2HPO4 were found to be the most important factors. Subsequently, response surface methodology was employed to determine the maximum nisin production at the optimum con
centration of key factors. The nisin production was found to correlate to the three parameters. The optimal values of the three parameters were determined as 2235 g/L yeast extract, 2480 g/L glucose and 1365 g/L K2HPO4. The nisin production was 2 157 IU/mL, and the actual experimental result was 2 115 IU/mL, which shows the results are reasonable.
