阿托伐他汀对家兔动脉粥样硬化模型PPARγ及MMP-9表达的影响

摘要: 目的:研究阿托伐他汀对动脉粥样硬化(AS)组织中过氧化物酶体增殖激活受体γ(PPARγ)和金属蛋白酶-9(MMP-9)表达的影响。方法:将实验动物家兔分为对照组(n=8)、高脂模型组(n=8)和阿托伐他汀干预组(n=8)。高脂模型组用高脂饲料饲养16周,阿托伐他汀干预组除高脂饲养外加用阿托伐他汀;应用免疫组织化学方法观察各组主动脉粥样硬化组织中PPARγ和MMP-9的表达。结果:高脂模型组兔主动脉壁所含PPARγ的阳性标记物百分比(14.38%±2.58%)明显高于对照组(7.82%±0.96%)(P<0.01),所含MMP-9的阳性标记物百分比(17.13%±1.58%)也明显高于对照组(2.72%±0.36%);阿托伐他汀干预组兔主动脉壁所含PPARγ的阳性标记物百分比(16.11%±2.35%)显著高于高脂模型组(P<0.01),而所含MMP-9的阳性标记物百分比(12.11%±2.05%)低于高脂模型组(P<0.01)。结论:阿托伐他汀增加动脉粥样硬化病变部位PPARγ的表达,抑制MMP-9的表达。

Abstract: To study the effects of atorvastatin on the expressions of peroxisome proliferators-activated receptor gamma(PPARγ)and matrix metalloproteinase-9(MMP-9)in atherosclerosis(AS) tissue. Methods Atherosclerotic rabbit models on aortas were established with the high-cholesterol diet,and animals were divided into control,model and atorvastatin groups(n=8).The expressions of PPARγ and MMP-9 and the effects of atorvastatin on them were observed by means of immunohistochemistry. Results The expression of PPARγ in model group(14.38%±2.58%)was higher than that in control group(7.82%±0.96%)(P<0.01),and the expression of MMP-9 in model group(17.13%±1.58%)was also higher than that in control group(2.72%±0.36%)(P<0.01).The expression of PPARγ in atorvastatin group(16.11%±2.35%)was higher than in model group(P<0.01),and the expression of MMP-9 in atorvastatin group(12.11%±2.05%)was lower than that in model group (P<0.01). Conclusion Atorvastatin can increase the expression of PPARγ and inhibit the expression of MMP-9.These results suggest that atorvastatin has anti-inflammatory effects which are probably related to its activating PPARγ and inhibiting MMP-9.
