MMP-9及其抑制因子TIMP-1在膀胱癌中的表达及意义

摘要: 目的:探讨基质金属蛋白酶9(MMP-9)及其抑制因子(TIMP-1)在膀胱移行细胞癌(TCCB)中的表达及意义。方法:应用免疫组织化学和RT-PCR方法检测47例TCCB组织和5例正常膀胱组织MMP-9和TIMP-1蛋白的表达情况。 结果:在正常膀胱组织中MMP-9、TIMP-1不表达或呈分布均匀的微量表达,而在TCCB中则有不同程度的阳性表达,二者表达量比较差异均有显著性(P<0.05)。随着TCCB临床病理分期、分级的升高,MMP-9的表达呈现增高趋势(P<0.01),但TIMP-1表达情况与肿瘤临床病理分期、分级无明显关联(P>0.05)。MMP-9和TIMP-1在初发TCCB和复发TCCB中的表达差异均无显著性(P>0.05)。结论:MMP-9在TCCB的发生、发展中具有促进作用;MMP-9和TIMP-1之间的平衡失调可能是肿瘤侵袭和转移的分子机制之一。

Abstract: Abstract:Objective To study the expressions and significances of matrix metalloproteinases-9 (MMP-9) and tissue -inhibitor of matrix metalloproteinase-1 (TIMP-1) in transitional cell carcinoma of bladder (TCCB).Methods Immunohistochemistry and RT-PCR were used to detect the expressions of MMP-9 and TIMP-1 in 47 surgical respected specimens from patients with TCCB (experimental group) and 5 normal bladder tissues (control group).Results MMP-9 and TIMP-1 had different degree expressions in experimental group,but the expressions were weak or negative in control group.Furthermore,the positive expression levels of MMP-9 were significantly increased with stages and grades (P<0.01); however the expression of TIMP-1 did not show significantly correlation with different grades and stages of TCCB tissue(P>0.05).Both MMP-9 and TIMP-1 had no significant correlation with first-occurrent TCCB and recrudescent TCCB.Conclusion MMP-9 plays advance roles in the occurrence and development of TCCB; the unbalance between MMP-9 and TIMP-1 is one of important molecular mechanisms in the aggressive and metastasis biologic behavior of tumor.
