实验性肺纤维化小鼠IL-1和TNF-α的动态表达及其意义

摘要: 目的:探讨博莱霉素致肺纤维化小鼠各时期支气管肺泡灌洗液(BALF)及肺组织中白细胞介素1(IL-1)和肿瘤坏死因子-α(TNF-α)的mRNA表达规律及其意义。方法:40只雄性昆明种小鼠,随机分为对照组(n=10)和实验组(n=30)。分别于生理盐水和博莱霉素(5 mg•kg-1)一次性气管内灌注后第3、7、14、28和56天行支气管肺泡灌洗并处死小鼠,取支气管肺泡灌洗液和肺组织,应用RT-PCR法测定其中的IL-1 mRNA和TNF-α mRNA相对转录水平。结果:①BALF中,实验组IL-1 mRNA和TNF-α mRNA表达均高于对照组(P<0.05),且实验组中两者的表达高峰均在第14天。②肺组织中,实验组IL-1 mRNA和TNF-α mRNA表达水平均高于对照组(P<0.01),且实验组中两者表达高峰分别在第7和14天,此后逐渐下降,但仍然保持高于正常水平表达。结论:IL-1 mRNA和TNF-α mRNA表达高峰时相均在肺纤维化发病早期,因此推断其在致肺纤维化损伤中发挥重要作用,并依IL-1 mRNA和TNF-α mRNA表达水平可判断病变发展进程和活动性。

Abstract: Abstract:Objective To investigate the gene expressions of IL-1 and TNF-α in bronchoalveolar lavage fluid(BALF) and lung tissues in mice with pulmonary fibrosis(PF) at various periods and to study the effects of cytokines on pulmonary fibrosis. Methods Forty male mice were randomly divided into two groups:experimental group(n=30) and control group(n=10), and poured respectively by bleomycin solutions and saline and killed on the 3rd, 7th,14th,28th and 56th day. RT-PCR method was used to analyse the mRNA expressions of IL-1 and TNF-α.Results ① The expressions of IL-1 mRNA and TNF-α mRNA in BALF in experimental group were higher than those in control group(P<0.05),and peaked on 14th day. ②The mRNA expressions of IL-1 and TNF-α in lung tissues in experimental group were higher than those in control group(P<0.01), and peaked on the 7th and 14th day. Conclusion The mRNA expression time of IL-1 and TNF-α are different, by which the progression and activities of PF can be judged. They play an important role in the stage of pulmonary injury in PF.
