糖基化终末产物对大鼠肾系膜细胞基质金属蛋白酶2活性和表达的影响

Effects of advanced glycation end products on matrix metalloproteinase-2 activity and expressionin rat mesangial cells

摘要: 目的:观察糖基化终末产物(AGEs)对大鼠肾小球系膜细胞基质金属蛋白酶2(MMP-2)活性和表达的影响。 方法:体外培养正常大鼠肾小球系膜细胞,糖化牛血清白蛋白作为刺激物,未经糖化的牛血清白蛋白及常规培养作为对照,分别用含100 mg•L-1 AGEs(AGEs组)、100 mg•L-1牛血清白蛋白(BSA组)和不含刺激物(DMEM组)的无血清DMEM培养基,作用于系膜细胞48 h后,酶谱法检测各组系膜细胞上清MMP-2活性,逆转录聚合酶链式反应(RT-PCR)检测系膜细胞MMP-2 mRNA的表达。 结果:与BSA组及DMEM组比较,AGEs组系膜细胞MMP-2 mRNA的表达明显下降(P<0.01),上清MMP-2活性明显降低(P<0.01)。 结论:AGEs能降低大鼠肾小球系膜细胞MMP-2的活性和表达。

Abstract: Objective To investigate the effects of advanced glycation end products (AGEs) on matrix metalloproteinase-2 (MMP-2) activity and expression in rat mesangial cells. Methods Rat mesangial cells were treated with 100 mg•L-1 AGE-modified bovine serum albumin(AGEs group)or native bovine serum albumin(BSA group)for 48 h. Normal mesangial cells without any treatment were as a control (DMEM group). MMP-2 activity and mRNA were analyzed by zymography analysis and reverse transcriptase polymerase chain reaction (RT-PCR), respectively. Results Both MMP-2 activity and mRNA expression of mesangial cells in AGEs group were reduced significantly, compared with BSA or DMEM group (P<0.01). Conclusion AGEs can reduce the activity and expression of MMP-2 in rat mesangial cells.
