TGFβ1在乳腺癌间质新生血管周细胞中的表达

Expression of TGFβ1 in pericytes of angiogenesisin breast cancer

摘要: 目的:研究转化生长因子β(TGFβ1)在乳腺癌周细胞中的表达情况。 方法:取50例手术切除乳腺癌新鲜标本,通过免疫组织化学染色、原位杂交技术,同时采用Western印迹法、RT-PCR进行蛋白和mRNA半定量分析,观察TGFβ1在乳腺癌中的表达情况。 结果:TGFβ1蛋白质可表达在新生血管周细胞的胞浆中,同时也可表达于乳腺癌癌细胞及新生血管内皮细胞;TGFβ1 mRNA主要表达在血管周细胞的胞浆中,极少数癌细胞呈弱阳性表达。通过Western印迹法进行蛋白定量分析发现,随着血管密度的提高,TGFβ1的蛋白增高(P<0.01)。RT-PCR结果显示,随着血管密度的提高,TGFβ1的mRNA增高(P<0.01)。结论:TGFβ1通过周细胞参与血管生成。

Abstract: Objective To investigate the expression of transforming growth factor beta 1 (TGFβ1) in pericytes of angiogenesis in breast cancer. Methods The expressions of TGFβ1 in 50 samples with breast cancer were observed by immunohistochemistry and in situ hybridization. Western blotting and semi quantitative RT-PCR were also used to detect protein and mRNA. Results TGFβ1 protein expressed in cancer cells, endothelium cells and pericytes of blood vessels. TGFβ1 mRNA mainly distributed in the cytoplasm of pericytes. Western blotting results showed that TGFβ1 expression enhanced with the increase of MVD (P<0.01). RT-PCR results showed that TGFβ1mRNA expression enhanced with the increase of MVD (P<0.01). Conclusion TGFβ1 can regulate the angiogenesis through pericytes.
