大鼠脂肪肝模型的建立

摘要: 目的:探讨脂肪肝动物模型的制作方法。 方法:用缺乏蛋氨酸和胆碱的饲料喂养大鼠,复制脂肪肝模型,并进一步研究门静脉压、肝重/体重比以及肝脏病理结构的变化。 结果:大鼠肝脏第14天出现较多的脂滴积累,第21天超过2/3以上的细胞内均出现脂滴,肝细胞形态不规则,肝窦变窄,不规则;门脉压较正常大鼠升高。体重和肝重均小于正常大鼠。 结论:用缺乏蛋氨酸和胆碱的饲料喂养大鼠复制脂肪肝模型,喂养时间短,价格低廉,是一种较好的脂肪肝模型制作方法。

Abstract: Objective To investigate the method to establish fatty liver model of rats. Methods The rats were fed on a diet deficient in choline and amino acid (methionine in chief) and fatty liver models were established successfully. The changes of rat portal vein pressure, ratio of liver weigh/body weigh, and pathological structure of liver were observed. Results Fourteen days later, much more fatty droplets were accumulated in the livers of rats, more than two thirds of liver cells had fatty droplets accumulation after twenty-one and twenty-eight days. The form of fatty liver cells were irregular, liver sinus became narrow and irregular; portal vein pressure of the fatty liver was higher than that of normal one, weights of the liver and body were both lighter than that of normal one. Conclusion This method is effective and economical to establish fatty liver model of rat in short time.
