蝎毒逆转人乳腺癌细胞株耐药性研究

Study on effect of BMK scorpion venom on reversalof multidrug resistance in MCF-7/ADM cell line

摘要: 目的:探讨东亚钳蝎毒(buthus martensii kirsch, BMK)是否能够逆转人乳腺癌细胞的多药耐药性(MDR)。 方法:采用MTT法对蝎毒和阿霉素(ADM)合用时肿瘤细胞的半数抑制率(IC50)进行检测。荧光分光光度法检测蝎毒对细胞内ADM浓度的影响。 结果:蝎毒不同组别(100~350 mg•L-1)能分别提高MCF-7/ADM细胞株对阿霉素的敏感性 (P<0.05;P<0.01)。蝎毒不同组别(100~200 mg•L-1)能使MCF-7/ADM细胞内ADM的浓度升高。 结论:蝎毒能部分逆转人乳腺癌耐药细胞对阿霉素的耐药性。

Abstract: Objective To discuss reversal of multidrug resistance (MDR) of scorpion venom of buthus martensii kirsch (BMK) in MCF-7/ADM cell line. Methods MTT colorimetric method was used to detect the IC50 of MCF-7/ADM cell line when BMK was used in combination with adriamycin (ADM). Fluorescence spectrophotometer method was applied to detect the effect of BMK scorpion venom on intracellular ADM concentration of MCF-7/ADM cell line. Results BMK in different groups (100~350 mg•L-1) enhanced the sensitivities of MCF-7/ADM cells to ADM (P<0.05;P<0.01). BMK in different groups (100~200 mg•L-1) could markedly increase the intracellular ADM concentration of MCF-7/ADM cell line. Conclusion The results suggest that BMK can partly reverse the MDR of MCF-7/ADM cell line to ADM.
