摘要: 目的:观察血小板源性生长因子B(PDGF-B)在损伤大鼠坐骨神经中的表达,旨在阐明其在末梢神经再生中的作用。 方法:分别构建大鼠坐骨神经切割伤和碾压伤模型,应用免疫组织化学S-P法检测PDGF-B的表达。结果:两类损伤模型中,损伤近端神经和再生轴突PDGF-B的表达均增强。两类损伤模型均可见损伤神经远端的雪旺氏细胞PDGF-B表达量明显增加,在碾压伤模型中,随着轴突-雪旺氏细胞关系的恢复,PDGF-B表达水平下降,并最终恢复正常;在切割伤模型中,二者关系不能恢复,PDGF-B减至极低水平。 结论:坐骨神经损伤后PDGF-B的表达增强在末梢神经再生中发挥重要作用。

Abstract: Objective To observe the role of platelet-derived growth factor-B (PDGF-B) in the regeneration of peripheral nerve by studying its expression on injured rat sciatic nerves. Methods The rat sciatic nerves cut and crush-injured models were established, the expressions of PDGF-B on both injured nerves were detected by immunohistochemistry technique. Results The expressions of PDGF-B were increased in proximal nerve segments and regenerating axons in both models. Soon after both types of injury, considerable amounts of PDGF-B accumulated in Schwann cells in distal segments of both models. With restoration of the axon-Schwann cells relationship in the crush model, levels of PDGF-B tended to decrease, eventually returned to normal. In the cut model the relationship can not be restored, the PDGF-B was decreased to a very low level. Conclusion The increased expression of PDGF-B after sciatic nerve injury may contribute to peripheral nerve regeneration.
