癌基因Stat3与非小细胞肺癌侵袭和转移的关系

摘要: 目的:观察癌基因Stat3、CyclinD1及增殖细胞核抗原(PCNA)在非小细胞肺癌(NSCLC)中的表达情况,并分析其在非小细胞肺癌发生发展中的作用。方法:应用免疫组织化学SP法检测45例 NSCLC中Stat3蛋白、CyclinD1蛋白及PCNA蛋白的表达。结果:45例NSCLC中32例Stat3蛋白阳性表达,阳性表达率为71.1%; NSCLC不同组织学分级、有无淋巴结转移Stat3的表达差异有显著性(P<0.05),肿瘤不同大小及不同组织学分型间表达差异无显著性(P>0.05);Stat3与CyclinD1蛋白及PCNA蛋白的表达呈正相关(r=0.531, P<0.01;r=0.618,P<0.01)。结论:Stat3在NSCLC中异常表达,并可能参与NSCLC中CyclinD1蛋白的调节,促进细胞增殖,Stat3可能促进NSCLC侵袭和转移。

Abstract: Objective To investigate the expressions of Stat3, CyclinD1 and proliferating cell nuclear antigen (PCNA) in non-small cell lung cancer (NSCLC),and analyze their relationships with carcinogenesis and progression of NSCLC. Methods SP immunohistochemistry was used to detect the expressions of Stat3, CyclinD1 and PCNA in 45 cases of NSCLC. Results The positive rate of Stat3 was 71.1 % in NSCLC. The expression intensities of Stat3 was not associated with different tumor sizes and histological type, but it was associated with the expression intensities of CyclinD1 and PCNA (r=0.531, P<0.01;r=0.618, P<0.01)and it was re lated to histological grade and lymph node metastasis(P<0.05).Conclusion The expression of Stat3 is abnormal in NSCLC. Stat3 may involve in regulation of CyclinD1 in NSCLC and accelerate proliferation of NSCLC, it may play an important role in generation of NSCLC.
