牛磺酸在大鼠肝星状细胞中的抗氧化作用

Anti-oxidative stress effects of Taurine on rat hepatic stellate cells

摘要: 目的:探讨牛磺酸在体外培养的大鼠肝星状细胞(HSC)的抗氧化作用。 方法:将新鲜分离的大鼠HSC分别在普通培养基、含25及50 mmol•L-1牛磺酸的培养基中孵育培养,检测三种培养基上清中脂质过氧化物(LPO)、羟脯氨酸的表达水平及三种培养基培养的HSC表达转化生长因子β1(TGF-β1)的水平。利用BrdU法检测牛磺酸对HSC增殖的影响。结果:HSC活化后,各培养液上清中LPO浓度均较活化前明显上升(P<0.05);在含25及50 mmol•L-1牛磺酸培养基中培养的HSC与普通培养基培养的HSC相比, LPO及羟脯氨酸的分泌水平明显下降(P<0.05);与普通培养基相比,两组牛磺酸培养基培养的HSC表达TGF-β1明显减少(P<0.05); 50 mmol•L-1牛磺酸可以明显抑制HSC的增殖(P<0.05)。 结论:牛磺酸在体外实验中可以有效抑制大鼠肝星状细胞的LPO及羟脯氨酸的合成,并显示出显著的抑制HSC增殖的作用,提示牛磺酸可能在抗肝纤维化治疗中作为抗氧化剂具有重要的治疗价值。

Abstract: Objective To investigate the anti-oxidative stress effects of Taurine on rat hepatic stellate cells (HSC) cultured in vitro. Methods The freshly isolated rat HSC was incubated with general culture mediums and mediums with 50 or 25 mmol•L-1 of Taurine,respectively, the levels of LPO and hydroxyproline in supernatant as well as the expressions of TGF-β1 by HSC in different mediums were evaluated. The effect of Taurine on proliferation of HSC was verified by BrdU method. Results With the activation of HSC, LPO and hydroxyproline expressed into three mediums were increased significantly(P<0.05).This up-regulation was suppressed significantly by administration of 50 mmol•L-1 or 25 mmol•L-1 Taurine (P<0.05); Meanwhile, compared with the general medium group, the expressions of TGF-β 1 mRNA were decreased dramatically in HSC incubated with 50 mmol•L-1 or 25 mmol•L-1 Taurine pre-treated mediums(P<0.05); furthermore, administration of 50 mmol•L-1 Taurine suppressed the proliferation of HSC significantly(P<0.05). Conclusion Taurine is effective in inhibiting the production of LPO as well as hydroxyproline by HSC and inhibiting proliferation of HSC, suggesting that Taurine, as an anti-oxidative stress factor, may have a potential therapeutic value on liver fibrosis.
