HSP65-MUC1/HSP65抗体免疫复合物对人树突状细胞的活化作用

Activation and maturation of dendritic cells induced by HSP65-MUC1/HSP65 immune complex

摘要: 目的:探讨免疫复合物能否诱导树突状细胞的活化与成熟。 方法:用免疫复合物刺激来自人外周血经细胞因子IL-4和GM-CSF诱导的未成熟的树突状细胞, 观察免疫复合物对未成熟树突状细胞的活化与成熟的影响。 结果:HSP65-MUC1/HSP65免疫复合物与HSP65-MUC1抗原或HSP65抗体相比较,能够显著地活化树突状细胞,使其表面的协同刺激分子CD86的表达显著上调,为激活细胞毒性T淋巴细胞提供第二信号,这一结果说明免疫复合物具有交叉递呈的作用;同时,加入免疫复合物的树突状细胞培养上清中IL-6和TNFα分泌量明显增加。结论: 免疫复合物能够诱导树突状细胞的活化与成熟,免疫复合物具有交叉递呈的作用。

Abstract: Objective To explore the activation and maturation of dendritic cells (DC) stimulated by immune complex. Methods Immune complex was added to the immature DC induced by IL-4 and GM-CSF from PBMC and the expressions of costimulatory molecules CD86 on the surface of DC and cytokines in the supernatant were detected. Results Compared with antigen or antibody, immune complex could significantly activate the DC by upregulating the expression of CD86 on the surface of DC which was the second signal for the activation of cytotoxic T lymphocytes and the secretions of IL-6 and TNFα. Conclusion Immune complex can induce the activation and maturation of DC.
