阴、阳离子交换示差组合色谱法的设计及在纤溶酶纯化中的应用

摘要: 目的:探索以阴、阳离子交换色谱串接组合为方式,进行高分辨率分离纯化蛋白质的可行性。方法:阴、阳离子交换示差组合色谱法,以等度洗脱方法进行的阴、阳离子交换组合色谱,以及亲和色谱的纯化结果相对比。结果:聚丙烯酰胺凝胶电泳考马斯亮蓝染色法结果显示,阴、阳离子交换示差组合色谱法,具有较好的分离纯化效果。结论:以等度洗脱、pH梯度与盐梯度洗脱串接组合的阴、阳离子交换示差组合色谱是一种较好的组合纯化的方式,达到亲和色谱的纯化效果。

Abstract: Objective To explore the possibility of achieving the highest resolution by using combination of anion and cation exchange chromatography. Methods Purification results were compared among the differential combination chromatography of anion and cation exchange, anion and cation exchange chromatography of both isocratic elution, and affinity chromatography. Results It was showed by PAGE that the differential combination chromatography of anion and cation exchange had much better purifying ability than anion and cation exchange chromatography of isocratic elution.Conclusion Differential combination chromatography of anion and cation exchange can get good result which can be got under the affinity chromatography conditions in the purification of proteins.
