人参合剂的抗诱变作用

Antimutagenic effect of traditional Chinese medicine mixture

摘要: 目的:探讨人参、五加皮、甘草及猪苓4味中药合剂对诱变剂导致染色体损伤的拮抗作用。方法:选择20例健康献血者为实验对象,分为正常对照组、博来霉素(BLM)诱变组、中药合剂组和中药合剂抗诱变组。利用诱变剂敏感性实验方法,所有实验对象均于实验当日常规采集外周血,肝素抗凝,将0.3 mL外周血接种于培养液中,37℃恒温培养箱中静置培养72 h。收获细胞及制玻片后,姬姆萨染液染色。检测BLM对正常人外周血淋巴细胞染色体的影响,观察以人参、五加皮、 甘草、猪苓4种中药的混合制剂作为染色体稳定剂的抗诱变作用。结果:BLM诱变组每细胞染色单体断裂(0.733±0.275) (b/c值)明显高于正常对照组( 0.018±0.076)(P<0.01),BLM诱变组(0.733±0.275)与中药合剂抗诱变组(0.566±0.761)比较b/c值差异有显著性(P<0.01)。结论:BLM导致正常人外周血淋巴细胞染色体的断裂率明显增高,人参、五加皮、甘草及猪苓4种中药合剂可以明显抑制BLM诱发的染色体断裂,提示此人参合剂可以作为染色体稳定剂,能够提高机体基因组稳定性。

Abstract: Objective To study the resistance effect of traditional Chinese medicine mixture to the damage of chromosome caused by Bleomycin (BLM) mutagen. Methods Twenty healthy blood donors were divided into four groups:control group,BLM induced group, traditional Chinese medicine mixture group and traditional Chinese medicine mixture anti-induced group. All blood samples were collected at the day of blood culture, hepalean preventing to coagulate. Culture medium were placed into incubator for 72 h after accepted 0.3 mL blood. The cells were harvested and microscope slides were prepared before Giemsa staining. The influence of BLM on the peripheral blood lymphocyte chromosome of the normal body was detected. The antimutagenic effects of the four-herb mixture including Ginseng,Cortex Acanthopanasia radicis,Liquorice,Grifola umbellate as chromosome stabilizer were observed. Results The b/c value in mutagenesis group (0.733±0.275) was obviously higher than that in control group (0.018±0.076)(P<0.01), the difference of b/c value between mutagenesis group (0.733±0.275) and traditional Chinese medicine mixture anti-induced group (0.566±0.761) was significant(P<0.01). Conclusion BLM can increase the fracture rate of the peripheral blood lymphocyte chromosomes in normal body obviously, the mixture including four traditional Chinese medicine can inhibit the fracture rate of chromo some induced by BLM obviously. It suggests that the mixture including four traditional Chinese medicine can improve the genome stability of the organism as chromosome stabilizer.
