摘要:

目的:探讨维甲酸-干扰素诱导死亡相关基因-19(GRIM-19)及其靶基因产物STAT3在胃癌组织中的表达及其临床意义,阐明其在胃癌发生和发展过程中的作用。方法:应用 RT- PCR、免疫组织化学染色法和Western blotting法检测48例胃癌组织、癌旁组织及正常胃组织中GRIM-19和STAT3的mRNA及蛋白表达水平,并分析其蛋白的表达情况与临床病理特征的关系。结果: GRIM-19 mRNA及蛋白表达水平在胃癌及癌旁组织较正常胃组织表达减弱( P<0.01),且GRIM-19蛋白表达水平与胃癌分化程度和临床分期有关联(P<0.01);STAT3 mRNA及蛋白表达水平在胃癌及癌旁组织较正常胃组织表达增强( P<0.01),且STAT3蛋白表达水平与胃癌分化程度、淋巴结转移及临床分期有关联(P<0.01);GRIM-19与STAT3在胃癌组织中表达水平呈负相关关系(r=-0.396,P<0.01)。结论:在胃癌组织中有STAT3持续高表达和GRIM-19低表达共存现象,可能与正常细胞发生恶性转化和异常增殖有关,促进了胃癌的发生和发
展。

Abstract:

Objective To study the expressions of GRIM-19 and its target gene product STAT3 in human gastric carcinoma(GC)tissues and their clinical  significances,and clarify their effects in occurrence and development of gastric carcinoma.  Methods  RT-PCR,immunohistochemistry dyeing and Western blotting were used to detect the mRNA and protein expressions of GRIM-19 and STAT3  in GC tissues,tissues surrounding carcinoma and normal gastric tisssues. The correlations between  the expressions of  GRIM-19 and STAT3 and  the  clinicopathologic characteristics of gastric carcinoma were analyzed statistically. Results The mRNA and protein levels of GRIM-19  in GC tissues as well as tissues surrounding carcinoma were obviously lower than those in normal tissues (P<0.01). Furthermore,the expression of GRIM-19 protein  was correlated to cell differentiation and clinical stage of GC (P<0.01). In contrast,the mRNA and protein levels of STAT3  as well as tissues surrounding carcinoma in GC tissues were higher  than those in normal tissues (P<0.01),and the expression of STAT3 was correlated to cell differentiation,lymphatic metastasis and clinical stage of GC(P<0.01). The significantly negative correlation was found between the expressions of GRIM-19 and STAT3 in GC tissues (r=-0.396,P<0.01). Conclusion
The low expression of GRIM-19 and the high expression of STAT3 co-exist in GC tissues. It may be related to malignant transformation and abnormal proliferation of cells,which facilitates the development of GC.
