应用几丁质胶原网架构建组织工程软骨

摘要: 目的:探索利用几丁质胶原网架构建组织工程人工软骨的方法。 方法:取2周龄的新生兔关节软骨,经消化、传代培养后,将获得的软骨细胞与牛Ⅰ型胶原、人血冻干纤维蛋白原、凝血酶按一定比例混合,将几丁质纤维网架浸入上述混合物中制成人工软骨培养物并在体外培养。将培养1周的组织工程培养物,植入同种异体动物皮下组织培养4周,观察组织工程软骨的形成情况。 结果:细胞在几丁质网架培养物中分布均匀,细胞多呈三角形或梭形,培养物体积收缩不明显,经体内培养存活的软骨细胞分泌Ⅱ型胶原,有血管和炎性细胞的侵入。结论:用胶原蛋白和人血纤维蛋白混合凝胶与几丁质纤维网架载体为支架植入软骨细胞以后,在体外和体内都可以培养构建出较大的组织工程软骨。

Abstract: Objective To explore the method to construct articular cartilage with chitin-collagen scaffold. Methods Chondrocytes were isolated from articular cartilage of 2-week-old rabbits,digested by collagenase Ⅱ, and subcultured to passage 2 in vitro. After digested, the cells were mixed with bovine I type collagen, human blood freeze-dry fibrinogen, and thrombosin according to a certain proportion and mixed into a kind of net-like carrier that was made of chitin. After cultured one week in vitro, the culture was implanted into subcutaneous tissue in the back of a allogenic adult rabbit for 4 weeks. Results After one week, the cells appeared in triangle or like shuttle, and well distributed in the scaffold. The volume was no change obviously. After cultured in vivo,the chondrocyte secreted collagen Ⅱ and had invaded to the vessels and cells of inflammation. Conclusion Chondrocytes cultured in collagenⅠ,fibrin and chitin mixture may be used as a scaffold that can make tissue engineering cartilage in vitro or in vivo.
