GLRA1基因多态性与精神分裂症的关系

Association between GLRA1 locus polymorphismin chromosome 5 and schizophrenia

摘要: 目的:探讨人类第5号染色体GLRA1基因多态性与精神分裂症的关系。方法:应用PCR-RFLP方法检测91个中国汉族精神分裂症患者及其健康父母双亲组成的核心家系单核苷酸多态性(SNP)rs1428157。 结果:父母组和患者组rs1428157等位基因频率的分布没有偏离Hardy-Weinberg平衡;传递不平衡检验(TDT)显示,杂合父母传递给受累子女与非传递等位基因频率分布差异无显著性;单倍型相对风险分析(HRR)显示,该基因位点与精神分裂症无关联;该基因位点与精神分裂症阳性症状的读心症和思维连贯性障碍密切相关。 结论:GLRA1位点的基因多态性与精神分裂症无关联,但其位点多态性与精神分裂症的读心症和思维连贯性障碍症状密切相关。

Abstract: Objective To investigate the genetic association between the GLRA1 locus in chromosome 5 with susceptibility to schizophrenia among 91 Chinese Han family trios consisting of fathers, mothers, and affected offsprings with schizophrenia. Methods Genomic DNA was isolated from the whole blood samples. The PCR-based restriction fragment length polymorphism (RFLP) technique was used to detect the single nucleotide polymorphism(SNPs),rs1428157, which was a base mutation for C to A at the GLRA1 locus. Results The distribution of genotype frequencies of rs1428157 was not deviated from the Hardy-Weinberg equilibrium; Haplotype relative risk test (HRR) did not show a genetic association between rs1428157 and schizophrenia (χ2=0.293,P>0.05); Transmission disequilibrium test (TDT) did not show significant biased transmission from parents to affected offspring(χ2=0.281,P>0.05); There were significant correlations between allele frequencies and incoherence of thinking and delusion of being revealed. Conclusion The rs1428157 itself is not associated with schizophrenia, but it has significant correlation with incoherence of thinking and delusion of being revealed.
