实验性哮喘小鼠T淋巴细胞对B细胞分泌特异性IgE的调节作用

Regulatory effect of T lymphocyte on specific-IgEsecreted by B lymphocyte in mouse with asthma

摘要: 目的:探讨支气管哮喘发病中T细胞对B细胞产生特异性IgE(SIgE)的影响。方法:建立小鼠支气管哮喘模型,分离小鼠脾脏T、B细胞,分组进行细胞培养,观察哮喘鼠T细胞对B细胞分泌SIgE的影响,并以ELISA法检测不同组合细胞培养上清中SIgE。结果:在有或没有特异性抗原参与时,哮喘鼠B细胞分泌SIgE水平均明显高于对照组B细胞(P<0.05);自身T细胞均可降低哮喘鼠B细胞分泌SIgE的水平(P<0.05);无论特异性抗原是否参与,从数值上看,哮喘鼠T细胞有使对照鼠B细胞SIgE分泌水平增加趋势,但差异无显著性(P>0.05)。 结论:T细胞在支气管哮喘的发病过程中,尤其是SIgE的产生中起重要的调节作用;T细胞功能异常是哮喘发病的原因之一。

Abstract: Objective To study the regulation and interference of T lymphocyte on antigen-specific IgE (SIgE) antibodies by B lymphocyte in bronchial asthma mouse model and clarify the role of T lymphocyte in the pathogenesis of asthma. Methods The asthma mouse model was set up at first, then T and B lymhphocytes were isolated from the mice spleen and were cultivated in different combinations. The supernates were collected and the specific anti-OVA antibody IgE in the supernate were tested with enzyme linked immunosorbent assay (ELISA). Results The level of SIgE secreted by B cells from asthma mouse was higher than that from the control mouse (P<0.05). After regulated by own T lymphocytes, the level of SIgE secreted by B cells from asthma mouse decreased significantly (P<0.05). With or without specific antigen, after regulated by T cells from asthma mouse, the level of SIgE secreted by B cells from the control mouse increased, but there was no significant difference (P>0.05). Conclusion T lymphocyte has a key role in regulating and controlling the pathogenesis of asthma. And T cell functional abnormality is one of the important mechanisms of asthma.
