乙型肝炎病毒相关性肝癌患者癌组织中HBx和CEACAM1的表达及其意义

Expressions of HBx and CEACAM1 in HBV-related hepatocellular carcinoma and their significances

摘要:

目的:检测乙型肝炎病毒x蛋白(HBx)和癌胚抗原相关细胞黏附分子1(CEACAM1)在乙型肝炎病毒相关性肝癌(乙肝相关性肝癌)组织中的表达,探讨二者在肝癌发生、发展及转移中的关系及意义。方法:采用免疫组织化学方法(SP法)检测81例乙肝相关性肝癌组织中HBx和CEACAM1的表达,分析HBx和CEACAM1与乙肝相关性肝癌临床病理特征的关系。Western blotting检测人正常肝细胞系QZG、肝癌细胞HepG2及稳定转染HBx的肝癌细胞HepG2(HepG2-X
)中CEACAM1的表达。结果:81例乙肝相关性肝癌组织中HBx表达阳性率为74.07% (60/81),CEACAM1表达阳性率为71.60% (58/81),二者与门静脉侵袭、淋巴结转移和TNM分期有关联(P<0.05,P<0.01)。HBx与CEACAM1的表达呈负相关(rs=-0.310,P<0.05);稳定转染HBx的肝癌细胞HepG2(HepG2-X)中, CEACAM1蛋白表达水平与肝癌细胞HepG2及人正常肝细胞系QZG比较显著降低。结论:HBx的高表达和CEACAM1的低表达与乙肝相关性
肝癌的侵袭、转移有关联,HBx可能通过抑制CEACAM1表达促进肿瘤的发生与发展。

Abstract:

Objective To investigate the expressions of hepatitis B virus X protein (HBx) and carcinoembryonic antigen-related cell adhesion molecule 1 (CEACAM1) in HBV-related hepatocellular carcinoma tissue and to explore the relationship between the two genes and occurrence,development and invasion of hepatocelular carcinoma.
Methods  The expressions of HBx and CEACAM1 in 81 cases of HBV-related hepatocellular carcinoma tissues were detected by immunohistochemistry method,and the relationship with clinicopathological features was analyzed. The expressions of CEACAM1 in the human normal hepatocyte cell line QZG, hepatocellular carcinoma HepG2 and HepG2-X(HepG2 cells transfected with HBx) were detected by Western blotting. Results The positive rates of HBx and CEACAM1 expressions in 81 cases of HBV-related hepatocellular carcinoma tissues were 74.07%(60/81) and 71.60%(58/81), respectively. The expressions of HBx and CEACAM1 were correlated with portal vein invasion,nodal metastasis and TNM staging(P<0.05 or P<0.01). The expression of HBx was negatively correlated with CEACAM1(rs=-0.310,P<0.05).The CEACAM1 protein was significantly  decreased in HepG2-X compared with HepG2 and human normal hepatocyte cell line QZG.Conclusion The high expression of HBx and low expression of CEACAM1 are correlated with the invasion and metastasis of HBV-related hepatocelular carcinoma,and HBx may promote HBV related hepatocarcinogenesis and development by       inhibition CEACAM1.
