银花泌炎灵片乙醇提取物对大肠埃希菌超微结构的影响及其抑菌作用机制

Influence of ethanol extract of Yinhuamiyanling Tablets in ultrastructure of Escherichia coli and 
its antibacterial mechanism

摘要:

目的:研究银花泌炎灵片95%乙醇提取物对大肠埃希菌标准菌株和临床耐药菌株的
体外抑菌活性,阐明其抑菌机制。方法:采用管碟法分别测定银花泌炎灵片95%乙醇提取物对大肠埃希菌标准菌株ATCC25922和18株泌尿系感染
产超广谱β-内酰胺酶(ESBLs)大肠埃希菌临床分离株的体外抑菌活性,测定其最小杀菌
浓度(MBC)。扫描和透射电子显微镜观察药物作用后菌体超微结构改变。结果:银花泌炎灵
片95%乙醇提取物对大肠埃希菌标准菌株ATCC25922和产ESBLs大肠埃希菌的MBC50和MB
C90分别为100 g/L-1和150~300 g/L-1。扫描电子显微镜下观察,经银花泌炎灵片95%乙醇提取物作用后大肠埃希菌菌体表面出现塌陷,两端可见凹陷和刺状突起,有的细菌严重变形;负染透射电
子显微镜下观察,药物作用后菌体折光性明显变差,菌体变形出现褶皱,且部分细胞壁出现
凹陷;超薄切片透射电子显微镜下观察,药物作用后菌体固缩膨大,细胞壁模糊不清甚至缺失,细胞质固缩解体,形成小空泡。结论:银花泌炎灵片95%乙醇提取物对大肠埃希菌标准菌株
ATCC25922和产ESBLs大肠埃希菌均有一定程度的抑制作用,其抑菌机制可能是药物干扰或破
坏大肠埃希菌细胞壁合成或破坏菌体内大分子量蛋白,以致菌体超微结构出现改变。

Abstract:

Objective To study the antibacterial activities of 95% ethanol extract
of Yinhuamiyanling Tablets in Eschericha coli  (E. coli) standard strains and clinical drug-resistant strains in vitro,and to clarify the antibacterial mechanism.
Methods Cylinder-plate method was used to test the antibacterial activities of Yinhuamiyanling Tablets of 95% ethanol extract on E.coli standard s
train ATCC25922 and 18 strains of urinary tract infection clinical E.coli producing extended spectrum β-lactamases (ESBLs),and the minimum bactericidal concentration (MBC) was determined.The bacterial ultrastructural changes were observed by scanning and transmission electron microscope before and after the drug application.Results The MBC50 and MBC90 of 95% ethanol extract of Yinhuamiyanling Tablets in  E.coli standard strain ATCC25922 and clinical E.coli producing ESBLs were 100 g/L-1 and 150-250 g/L-1.The E.coli cells showed surface collapse,depression and thorn-like protuberances at both ends,severely deformed under  scanning electron microscope after treated with 95% ethanol extract of Yinhuamiyanling Tablets;negative staining revealed that the bacterial refraction deteriorated under transmission electron microscope,bacteria deformation appeared to fold,and part of
the cell wall were depression;the negative staining  transmission electron microscope results  showed  bacteria shrinkage,swelling,the cell wall blurred or even
 missed,and the cytoplasm was pyknotic and disintegration to small vacuoles.Conclusion 95% ethanol extract of Yinhuamiyanling Tablets  has a certain degree of inhibitory effect on E.coli standard strain ATCC25922 and E.coli producing ESBLs,and its mechanism may be related to its interference or disruption of the E. coli
cell wall synthesis or destruction of large molecular proteins,which results in  the ultrastructural changes of E.coli.
