锌和铬对糖尿病小鼠的降血糖及抗氧化作用

摘要: 目的:观察锌(Zn)、铬(Cr)对实验性糖尿病小鼠的降血糖和抗氧化作用。方法:用四氧嘧啶按200 mg·kg-1剂量左下腹腔内1次性注射制作糖尿病小鼠模型。动物随机分为4组:锌组、铬组分别按5.1 mg·kg-1·d-1(Zn2+)和40 μg·kg-1·d-1(Cr3+)灌服硫酸锌和三氯化铬溶液,正常对照组和实验对照组灌服相应体积的蒸馏水。实验周期4周。葡萄糖氧化酶法测血糖,黄嘌呤氧化酶法测肝组织超氧化物歧化酶(SOD),比色法测定肝组织中谷胱甘肽过氧化物酶(GSH-Px )活力和总抗氧化能力(T-AOC),硫代巴比妥酸法测定丙二醛(MDA)水平。结果:锌组、铬组血糖水平低于实验对照组(P<0.01),SOD、GSH-Px活性及T-AOC 水平高于实验对照组(P<0.05或P<0.01)且低于正常对照组(P<0.01),MDA水平明显降低(P<0.01)。 结论:锌、铬具有降低实验性糖尿病小鼠血糖,增强其抗氧化能力的作用。    

Abstract: Objective To study the effects of zinc and chromium on declining blood glucose level and improving antioxidant activity in diabetic mice. Methods The models of experimental diabetic mice were established by abdominal cavity injection of alloxan with the dose of 200 mg·kg-1 . The mice were divided into 4 groups: zinc, chromium, normal control and experimental control group. The mice in zinc and chromium groups were respectively administered orally with ZnSO4 and CrCl3 solution with the dose of 5.1 mg·kg -1·d-1(Zn2+) and 40 μg·kg-1·d-1(Cr3+). Xanthine-oxidasemeasurement method was used to measure the quantity of SOD in hepatic tissues. Colorimetric method was used to measure the activities of GSH-Px and T-AOC in hepatic tissues. Thio-barbituric acid was used to measure the level of MDA. After four weeks, blood glucose, T-AOC, MDA, SOD, GSH-PX were detected. Results Compared with experimental group, blood glucose levels in zinc and chromium groups were decreased(P<0.01), the activities of SOD and GSH-Px as well as T-AOC were increased(P<0.05 or P<0.01). Meanwhile, the level of MDA was declined remarkably(P<01). Compared with normal control group, the activities of SOD, GSH-Px and T-AOC were decreased (P<0.01). Conclusion Zinc and chromium can decrease the blood glucose and improve the antioxidant activity in diabetic mice.
