TGF-β1对大鼠肾小球系膜细胞PAI-1表达的影响

Effects of TGF-β1 on expression of PAI-1 in rat mes angial cells

摘要: 目的:观察不同浓度重组人转化生长因子-β1(TGF-β1)对大鼠肾系膜细胞增殖情况和纤溶酶原激活物(PAI-1)抑制物表达水平的影响。方法:采用MTT和流式细胞仪检测细胞增殖,Western blotting和RT-PCR法检测PAI-1的表达水平。结果:与对照组相比,0.1和0.5 μg·L-1 TGF-β1诱导肾小球系膜细胞增殖,增殖率分别为(146.6±10.2)%和(134.6±14.5)%,PAI-1 mRNA和蛋白表达增强;而5和25 μg·L-1 TGF-β1诱导的细胞增殖率为(75.2±13.2)%和(67.5±11.1)%,PAI-1 mRNA和蛋白表达无明显变化。结论:低剂量TGF-β1促进大鼠系膜细胞增殖、诱导PAI-1表达;高剂量TGF-β1则抑制系膜细胞增殖。

Abstract: Objective To study the effects of transforming growth factor-β1 (TGF-β1) on mesangial cells growth and expression of plasminogen activator inhibitor-1(PAI-1) in cultured rat mesangial cells stimulated by different levels of human recombinant TGF-β1. Methods The proliferation of mesangial cells was determined with MTT assay and FACS analysis. The levels of PAI-1 protein and mRNA were analyzed with Western blotting analysis and RT-PCR respectively. Results Compared with control group, 0.1 and 0.5 μg·L-1 TGF-β1 stimulated mesangial cells proliferation, the proliferation rate of mesangial cell was (146.6±10.2)% and (134.6±14.5)% respectively, and the expressions of PAI-1 mRNA and protein also increased (P<0.05). The proliferation rate was (75.2±13.2%) and (67.5±11.1)% in the group treated with 5 and 25 μg·L-1 TGF-β1, the expressions of PAI-1 mRNA and protein were constant. Conclusion Low level of TGF-β1 can trigger the proliferation of rat mesangial cells, induce the expression of PAI-1, and high level of TGF-β1 can inhibit the proliferation of mesangial cells.
