脐带血中血管内皮前期细胞的分离

摘要: 目的:从脐带血中分离单个核细胞以获得血管内皮前期细胞,并观察其生物学行为。方法:用密度为1.077的淋巴细胞分离液分离脐带血中单个核细胞,种植在铺有纤维连接素的培养板中,3 d后去除未附着细胞,观察附着细胞的生长情况,采用免疫组化法进行鉴定。结果:单个核细胞培养3 d后可见大量的附着细胞和少量梭形细胞,梭形细胞数量在第7天时达到高峰。附着细胞对第八因子相关抗原呈阳性反应。结论:在纤维连接素上培养来源于脐带血的单个核细胞可获得血管内皮前期细胞。

Abstract: Objective To isolate endothelial progenitor cells from umbilical cord blood and observe their biologic behaviors. Methods The mononuclear cells were isolated from umbilical cord blood using Ficoll gradients, and then were cultured on fibronectin. The characteristics of the attached cells were got and they were subjected to immunocytochemical method to analyze the expression of von Willebrand factor (vWF). Results After the mononuclear cells were cultured for 3 days, a large amount of attached cells and some spindle-shaped cells were observed. The number of spindle-shaped cells increased until 7th day. The immunocytochemical method showed that the attached cells were positively stained for vWF. Conclusion The endothelial progenitor cells may be isolated from umbilical cord blood.
